一种蛋白质药物载体复合微粒子的制备研究

摘要: 为了探索一种可携带药物的功能粒子的制备方法, 对以藻酸-钙水凝胶作载体, 复合磁性粒子和羟基磷灰石的纳米粒子生成纳米级复合微粒子进行了研究, 利用BCA 蛋白质量浓度测定法检测了复合微粒子对血红素c 的吸附能力。结果表明, 借助于水凝胶在反相胶束微水池中快速胶联, 铁磁性纳米粒子直接被包埋在水凝胶的胶联骨架内, 羟基磷灰石纳米粒子以水凝胶为模板共沉生成并镶嵌在水凝胶骨架中, 反相胶束水相中的快速凝胶化有效地制约了复合粒子的大小。吸附结果表明, 制备得到的复合微粒子对血红素c 具有明显的亲和性, 其主要原因是载体微粒子复合了羟基磷灰石。

Abstract: In order to explore a method to preparing a sort of functional composite particles with controllability and carrying behavior, composite submicron particles of hydroxyapatite and magnetite were prepared by microemulsion method using alginate-calcium hydrogel as carrier, and the affinity of the obtained composite particles to cytochrome c was examined by BCA protein assay. The results show that the magnetite nanoparticles are entrapped into the crosslinked network of alginate-calcium gel, the hydroxyapatite particles co-precipitated are embedded inside the network, and the average particle size of the composite particles is 100 nm. The prepared composite particles shows a remarkable affinity to cytochrome c in aqueous solution due to the incorporation of hydroxyapatite.
