门多萨假单胞菌DS04灢T对PHBV降解性能研究

The Degradation of PHBV by [WTBX]Pseudomonas Mendocina[WTBZ] DS04T

摘要: 以3灢羟基丁酸灢co灢3灢羟基戊酸共聚物(PHBV)为唯一碳源培养PHBV降解菌DS04灢T,考察培养时间、
温度、培养基起始pH、摇床转速、接种体积分数以及装液量等环境因素对菌株产生PHBV 降解酶的影响,进而考察
菌株对PHBV的降解能力。并结合正交试验优选适宜的产酶条件如下:培养温度28 曟;培养时间32h;摇床转速
150r/min;装液量100 mL 培养基;培养基起始pH 为7.5;接种体积分数1.5%。在此优化条件下菌株产生的
PHBV降解酶有较高的活力,达(18.9暲1.2)U/mL。

Abstract: Using poly(3hydroxybutyrateco3hydroxyvalerate) (PHBV) as sole carbon source, the biodegradation of the PHBV by [WTBX]Pseudomonas mendocina[WTBZ] DS04T was studied. The effect of incubation time, cultivation temperature, initial pH, shaking revolution, liquid medium volume and inoculation content on PHBV degradability were studied. According to the results of orthogonal test, the optimal conditions of enzyme production were chosen as follows: temperature 28 ℃, incubation time 32 h, initial pH 7.5, shaking revolution 150 r/min, inoculation content 1.5% , and 100 mL culture medium is load in 250 mL flask. Under the optimum conditions, the PHBVdegrading enzymatic activity reached (18.9±1.2) U/mL.
