青霉T24-2降解甘蔗渣及发酵产氢的研究

Degradation of Sugarcane Bagasse by Penicillium sp.T24-2 and Hydrogen Production From Bagasse Hydrolysate

摘要: 通过单因素试验及正交试验对青霉(Penicil lium sp.)T24 -2 产纤维素酶及酶解甘蔗渣的条件进行优化, 并分析了产酸克雷伯氏菌(K lebsiella oxy toca)HP1 利用甘蔗渣糖化液产氢的特性。单因素试验结果表明, 蔗渣/ 麸皮质量比、曲酶/蔗渣质量比、固(曲酶+蔗渣)/ 液(水)质量比、糖化时间是影响产纤维素酶和蔗渣糖化率的主要因素。经正交试验并考虑大规模应用时的成本, 确定青霉T24-2 固态发酵产纤维素酶的最佳蔗渣/麸皮质量比为4∶6 。利用曲酶对甘蔗渣进行糖化的最佳条件为:曲酶/ 蔗渣质量比为1 ∶3, 固(曲酶+蔗渣)/液(水)质量比为 1∶3 , 糖化时间为20 h , 在此条件下甘蔗渣糖化率可达43.6%。产氢试验表明产酸克雷伯氏菌H P1 利用甘蔗渣糖化液, 每克甘蔗渣产氢可达41.0 m L。研究表明青霉T24 -2 是一株较好的产纤维素酶菌株, 可应用于纤维素生物质的开发与利用。

Abstract: The cellulase production by Penicillium sp. T24-2 and saccharification of sugarcane bagasse were studied by single factor test and orthogonal test. And the character of sugarcane bagasse produced hydrogen by Klebsiella oxytoca HP1 was researched. Single factor test results show that the mass ratio of bagasse to bran, starter material to bagasse, solid material to water and the saccharification time are the main factors affecting the saccharification. By orthogonal test, the optimal mass ratio of bagasse to bran is achieved as 4∶6. For the saccharification, the optimal conditions were obtained as: the mass ratio of starter material to bagasse is 1∶3, the mass ratio of solid material to water is 1∶3. Under above conditions, a saccharification yield of 43.6% after 20 h hydrolysis was obtained. When the saccharification liquid was used for hydrogen production by Klebsiella oxytoca HP1, a yield of 41.0 mL H2/g-bagasse was acquired. The results show that the Penicillium sp. T24-2 is a preferable strain for cellulase production and saccharification of cellulose biomass.
